Bacteria play an important role in wastwater treatment as they decompose
complex materials in simplest form.All life forms rely on bacterial processes for
their survival. Bacterial diversity is greater than the diversity of any other
organisms because bacteria play an important role in metabolic functions that
affect soil, plant and human health. The objective of this study was to determine
bacterial diversity in anaerobic complete mix bioreactor for treatment of
synthetic wastewater. Sludge samples from I-9 sewage treatment plant was used
as seed in the reactor.In the present study different bacterial species were
identified from isolation and identification.In this study different techniques were
used for the isolation and identification of bacteria. Those techniques were
colony morphology, and biochemical test were performed such as Gram staining,
catalase test, citrate test ,oxidase test .The result showed that different bacterial
species were isolated with respect to the colony morphology most colonies were
circular. paired, irregular, filamentous.
For identification of species the isolated and purifying colonies were shipped to
Microgen for characterization and amplification of 16sRNA gene sequencing and
the dominants species are Acinetobacter, salmonella, Aeromonas spp,
Citrobacter. Clostridium , Salmonella sp, Bacillus Clostridiumsp and uncultured
bacteria. This study play an important role in identification and isolation of
bacterial communities which are involved in treatment of domestic wastewater
and the effect of mixing speed on microbial profiling and also phylogenetic
analysis of microbes which are playing an important role in wastewater treatment
anaerobically. |
